چکیده

Background Recently, the role of long noncoding RNA (lncRNA) in rheumatic immune diseases has attracted widespread attention 1 . However, knowledge lncRNA connective tissue disease-associated interstitial lung disease (CTD-ILD) is limited. Objectives To explore expression profile peripheral blood mononuclear cells (PBMCs) CTD-ILD patients and possible mechanisms significantly differentially expressed involved CTD-ILD, especially systemic sclerosis (SSc)-ILD rheumatoid arthritis (RA)-ILD. Methods LncRNA microarray analysis was used to identify pattern dysregulation between without associated (CTD-NILD). Differential genes were identified by bioinformatic analysis. Relative levels five lncRNAs 120 SSc RA with or ILD detected quantitative reverse-transcription PCR (qRT-PCR). Results The differential gene revealed 46 upregulated while 194 downregulated group compared CTD-NILD (Figure 1). Gene Ontology Kyoto Encyclopedia Genes Genomes pathway analyses several significant biological processes signaling pathways, including NF-kappa B pathway, IL-17 Toll-like receptor cell pathway. QRT-PCR confirmed that selected target different groups. In particular, ENST00000604692 level higher than NILD (p<0.05, Figure 1); T311354 arginase-1were group; Furthermore, area under receiver operating characteristic curve for predicting from 0.797 Conclusion This research demonstrated, first time, specific PBMCs potential signal pathways related pathogenesis CTD-ILD. can effectively distinguish group, which may be a diagnostic indicator SSc-ILD RA-ILD. Interstitial lung disease in the connective tissue diseases.

The connective tissue diseases (CTDs) are inflammatory, immune-mediated disorders in which interstitial lung disease (ILD) is common and clinically important. Interstitial lung disease may be the first manifestation of a CTD in a previously healthy patient. CTD-associated ILD frequently presents with the gradual onset of cough and dyspnea, although rarely may present with fulminant respiratory ...

Interstitial lung disease in connective tissue disorders.

Some of the most pressing challenges associated with interstitial lung disease (ILD) are how best to define, diagnose, and treat connective tissue disease-associated ILD (CTD-ILD)--disorders with potentially substantial morbidity and mortality. In this focused review, we address aspects of prognosis for CTD-ILD and what indices might predict outcome, together with lessons that can be learnt fro...

Association between Long Noncoding RNA ANRIL Expression Variants and Susceptibility to Coronary Artery Disease

Animal cells possess thousands of long non-coding (lnc) RNAs, such as antisense noncoding RNA in the INK4 locus (ANRIL) , which have regulatory roles in the cells’ molecular mechanisms, including X-chromosome inactivation, and developmental processes.  These lnc RNAs are known to influence the extensive spectrum of age-related disorders. Accordingly, there is evidence for the role of...

Clinical Experience with Pirfenidone in Connective Tissue Diseases Related Interstitial Lung Diseases

Objective: Pirfenidone is a new, anti-fibrotic drug used for the treatmentof idiopathic pulmonary fibrosis (IPF). This study was designed to verify the effect of pirfenidone administrated in connective tissue diseases related interstitial lung disease (CTD-ILD). Methods: Twenty two patients diagnosed with CTD-ILD were administrated with a 6-months treatment with or without pirfenidone. At basel...

Lung cancer development in patients with connective tissue disease–related interstitial lung disease

Previous studies have reported that patients with idiopathic pulmonary fibrosis occasionally develop lung cancer (LC). However, in connective tissue disease (CTD)-related interstitial lung disease (ILD), there are few data regarding the LC development. The aim of the present study was to evaluate the clinical significance of LC development in patients with CTD-ILD. A retrospective review of our...
